Expertise: The lab of Wibke Diederich has worked on structure-based design and targeted synthesis of low molecular inhibitors for viral and parasitic proteases for several years. The main effort of their work is the targeted synthesis of small, target-specifically cut substance libraries with effective reactions of classic and modern organic synthesis chemistry. Inhibitor optimization occurs in repeating cycles from computer-supported molecule modelling, target-oriented synthesis, biological testing, and structure determination with regard to their affinity and pharmacokinetic characteristics. Established methods include not only synthetic expertise but also the recombinant expression of applicable proteins, biochemical characterization, as well as crystallization and structural elucidation of enzyme-ligand complexes via x-ray diffraction analysis.
