Expertise: The lab of John Ziebuhr works with the molecular biology of coronaviruses (including SARS and MERS) and related positive-sense RNA viruses. The focus of research is on the biochemical and functional characterization of virus-encoded enzymes that take part in viral RNA synthesis and/or processing (polymerases, helicases, exo- and endonucleases, methyltransferases, proteases) or modify host cell functions (macrodomain proteins, methyltransferases, phosphodiesterases). Numerous established molecular biological, biochemical, and reverse genetic methods, as well as established collaborations with structural biologists, support the work, which has led to the discovery of a series of enzymes that are only conserved in coronaviruses and a few related viruses; this has led to interesting points of approach for virus-specific antiviral therapies.
