Instruments

 

 

 

Our setup is a dual 2-photon system, consisting of an inverted and an upright setup. Both systems can be operated individually as confocal microscopes while they can share a multi-photon laser from SpectraPhysics.

 


 Inverted microscope

The inverted system has an Acousto-Optical Beam Splitter (AOBS), with a z-galvo stage and a resonant scanner, offering the following:

Confocal Laser Scanning Microscopy

    • Lasers:
      • 405nm
      • Argon laser (458nm, 476nm, 488nm, 496nm and 514nm)
      • White Light Laser (470nm-670nm)
    • Detectors:
      • 1 Transmitted Light PMT
      • 1 PMT
      • 3 Hybrid Detectors

Multi-photon Laser Scanning Microscopy

    • Laser:
      • SpectraPhysics Insight X3 with a 1045nm fixed line and a tuned line from 680nm up to 1300nm.
    • Detectors:
      • 4-Tune Mutli-photon module with 2 PMTs and 2HyDs

Digital Light Sheet

The inverted microscope is also equipped with a light sheet module suitable for small volumes (width up to 5mm).

        • 1.6x air, 2.5x air, 5x air, 10x air, 20x air
        • 25xWater, 40xWater, 63xWater (with motor correction collar)
        • 63xOil
        • 5x water diving objective for light sheet
        • 10x multi-immersion diving objective for light sheet
        • 5mm water, 7.8mm water/Glyc/BABB TwinFlect mirrors

Live Imaging Setup

The inverted microscope is also equipped with a temperature controller as well as a gas mixer for CO2, providing the option for live cell imaging.

 

 


 

Upright Microscope

The upright microscope is equipped with a Scientifica XY stage and a resonant scanner for fast imaging and can offer the following:

      • 638nm
    • Detectors:
      • 3 PMT
      • 1 Hybrid Detector

Multi-photon Laser Scanning Microscopy

    • Laser:
