Analytik von Anthocyanen und Metaboliten in Rohstoffen, Extrakten und biologischen Flüssigkeiten

Aims

The aim of this workpackage is the sensitive and specific analysis of anthocyanins and their metabolites in complex biological samples. This includes urine and plasma from animal and human studies as well as supernatants from cell culture experiments. Liquid chromatography in combination with mass spectrometric methods is used to optimize the identification and quantification of the main anthocyanins and selected metabolites. A strategy to handle and analyze a large number of samples and analysis results will be developed.

 

Results

Fast separation by ‘rapid separation LC’ at pressures up to 800 bars coupled to mass spectrometers with high mass accuracy allows for efficient identification and quantification of anthocyanins and metabolites. Simultaneous measurements in positive and negative ion mode enable the detection of e.g. glucuronated Anthocyanins and phenolic acids in a single LC run. Exact mass measurements allowed the differentiation of compounds which have almost identical molecular mass such as cumaroyl and rutinosid derivatives. Identification was confirmed by MS/MS experiments. In addition to conventional LC-MS measurements, mass spectrometry imaging for spatially resolved information and ambient ionization techniques for real-time analysis are employed for selected samples.
