The progress of the project "Signal-mediated chromatin changes in right heart failure" will be presented by project leaders Prof. Dr. med. Susanne Rohrbach, Institute of Physiology at JLU, and Prof. Dr. med. Michael Kracht, Rudolf Buchheim Institute of Pharmacology at JLU.
Pulmonary artery banding (PAB) in weanling rats will be used to study the transition from compensated (adaptive) right ventricular (RV) hypertrophy to RV failure. We will systematically characterize the regulatory non-coding genome, chromatin factors, histone modifications and their signal-mediated changes as major determinants of tissue-specific but also dynamic gene transcription in RV remodeling. These results will not only annotate the rat right and left heart transcriptome in relation to the regulatory genome but will also reveal how (re)organization of the enhancer landscape contributes to RV pathology and if epigenetic mechanisms offer for novel therapeutic interventions.
