Molecular mechanisms of CTCF mediated gene regulation in Drosophila
Insulation of chromatin domains from flanking regulatory activities is mediated by CTCF
and is fundamental for genome function. We have found that in Drososphila dCTCF is
required for development and that dCTCF is associated with CP190. This factor was
detected at CTCF, su(Hw) and GAF binding sites, at borders of H3K27me3 “islands”
and, unexpectedly, at the transcriptional start sites of actively transcribed genes. To
address the molecular mechanisms of CTCF function, we will analyze nucleosome
positioning, cofactor interaction and three-dimensional chromatin arrangements.
