Description
The Multiscale Imaging Platform is a core light microscopy facility of the Medical Faculty of the Justus-Liebig-University Gießen, located at the Center for Infection and Genomics of the Lung (CIGL). It serves as a core microscopy facility for the members of the Institute for Lung Health (ILH), as well as the Cardio Pulmonary Institute (CPI). Additionally, the facility is open to all interested research teams of the JLU or external institutions. Our core platform offers support, training, and on-request services in all steps of microscopy: experimental design, sample preparation, image acquisition, image data management (according to the FAIR principles), and image post-processing and quantification.
Instrumentation/Software
- Inverted Confocal / 2Photon: Leica SP8 DIVE with a White Light Laser (WLL), an Argon Laser, and a Spectra Physics Insight X3+ laser equipped with live imaging setup.
- Upright Confocal and 2Photon: Leica DM6 DIVE with 4 visible lasers (405nm, 488nm, 555nm, 638nm) and a Spectra Physics Insight X3+ laser.
- Slide scanner: Olympus VS200 for bright/dark-field, polarization and fluorescence microscopy. Fluorescence filters for DAPI, FITC, Cy3, Cy5, Cy7 (or similar), magnification objectives 4x, 10x, 20x, 40x and 100x, total capacity of 180 standard-size slides
- Image Analysis Server: Dual processor (48 cores in total), 1TB RAM, 3x nVidia RTX A5000 GPU, 200TB local storage.
- OMERO Image Data Management Software.
- Huygens Deconvolution Software (SVI): Software for offline image restoration and deconvolution. Suitable for confocal, and wide-field images.
- LAS X 3.5 with Lightning and 3D module: Computational clearing and 3D reconstruction and rendering.
- Ilastik: Image Classification and Segmentation with machine learning.
- QuPath: Bio-Image Analysis and Digital Pathology.
- ImageJ / Fiji with custom made plugins.
