Project B12 (new)
Molecular characterization of interactions between bone-substitute material and surrounding tissue in sheep, rat, and myeloma-mouse-model by transcriptome profiling using next generation RNA-Sequencing and functional validation
In project B12, for the first time, molecular changes through materials that have been incorporated in our murine, ovine, as well as the myeloma mouse model will be analyzed using high-throughput screening techniques. For the latter, RNA-sequencing will be performed, which we established for 1ng RNA. For validation purposes, a novel technique is applied using “human” DNA-microarrays for the investigation of e.g. ovine samples, and data will be functionally validated. This allows i) an integrative interpretation of the material-tissue-interaction (M1-M9, B10, T2) and ii)
These data will provide insight into the molecular changes caused by the incorporated materials and complement the histological (project T2) and imaging analyses (projects B8 and B9), as well as the in vitro generated results (projects B1 and B10).
